SPRINGLE
The SPRINGLE is the entry-level single channel surface plasmon resonance instrument. It provides high performance SPR measurements at a convenient price.
The SPRINGLE is ideal for fundamental research and development, is standard supplied with the special cuvette designed for biosensor and chemistry
research.
TWINGLE
The TWINGLE is designed for high quality, high accuracy measurements and is the economical choice for dual channel surface plasmon resonance measurements.
The reference channel data can be used to correct for experimental errors and/or matrix effects. The TWINGLE is standard supplied with a normal cuvette,
an electrochemical cuvette is optional.
ESPRIT
The ESPRIT is designed for high throughput SPR experiments. The automation allows reproducible measurements with the possibility to analyze up to 192
samples without user intervention.
The double channel reference measurement can be used to correct for systematic experimental errors and/or modified surfaces matrix effects.
Surface Plasmon Resonance
The KEI SPR instruments help the scientist to analyze bimolecular interactions in real time, without labelling requirements. Surface plasmon resonance (SPR) biosensors have become standard tools in life sciences, pharmaceutical research and material science laboratories. The study and characterization of molecular interactions are essential to explore the structure function relationships of bi-molecules and to aid our understanding of biological systems in life sciences.
With its open design the KEI SPR can be connected to any of our Autolab potentionstat/galvanostat instruments to perform electrochemical SPR measurements (ESPR). Together with a flexible software package, a wide range of applications can be accomplished.
Surface plasmon resonance can provide:

● Binding strength for molecular interactions - Affinity constant
● Discrimination between active and non-active molecules
● A qualitative tool for ligand screening

The KEI SPR instruments distinguish themselves by high performance and accuracy for a low total cost of ownership.
Cuvettes
The SPR instruments have a flexible open cuvette system. The cuvette has many advantages:
● Sample volume independent of application
● Incubation time independent of sample volume
● Sample incubation for 1 minute, 1 hour, 1 day or even 1 week
● Only the needle tip is in contact point with sample
● Viscosity independent, no clogging
● Sample recovery possible (for ESPRIT)
● Easy access for manual additions
The normal cuvette is the appropriate choice for measuring the lowest affinity binding systems as well as the highest affinity binding systems. The normal cuvette is standard supplied with the ESPRIT and TWINGLE.
A unique electrochemical cuvette which allows measurements on a 3-electrodes system is standard supplied with the SPRINGLE:
● Fixed contact point to the gold layer of the sensor disk (working
electrode)
● Reference electrode, which is replaceable
● Counter electrode is a fixed Platinum electrode
Software
All SPR instruments come with two software packages: the data acquisition software and the kinetic evaluation package.
The data acquisition software contains all necessary tools to control the instrument. Automatic control is done through measurement sequences. A sequence is a list of commands that is used to automatically control the instrument. The instrument comes with a standard set, ready-to-use sequences. It is also possible to create new sequences or to modify the existing ones to your specific needs.
The autosampler control window is used to select and identify the sample positions on the microwell plate, to set the incubation times and to activate a specific sequence/experiment.
The binding curve plot window shows the sensorgram real-time. In case of the ESPRIT and the TWINGLE both channels, the cuvette temperature and differential data of the association, dissociation and regeneration phase of the experiment are shown.
The users can add markers to any data point on the plot so that comments on special events or specific areas of interest can be marked and logged into an event recorder for later use.
Electrochemistry and SPR
Combined electrochemistry and SPR measurements can be performed using the Autolab NOVA software, where the users can record the SPR signal as a second signal while performing an electrochemical measurement. ESPR can be used to study e.g. electropolymerization and modified electrode surfaces.
Kinetic evaluation software
The measurement data collected by the data acquisition software can be analyzed with the kinetic evaluation software.
With the kinetic evaluation software the following parameters can be determined:
● Kinetic rate constants
● Affinity constants
● Thermodynamic parameters
The above parameters can be calculated using the multiple curves measured with different concentrations. The actual curves
can be fitted to different kinetic models. A wizard in the software assists in finding the right model and the proper initial parameters. A residual plot is available to verify the fitted data against the measured curve.
